The comorbidity of Alzheimer’s disease (AD) and age-related macular degeneration (AMD) has been established through clinical and genetic studies, with growing interest in identifying shared environmental factors. Leveraging recent advancements in record linkage techniques, a knowledge graph was constructed from literature to include significant risk factors for both conditions. An environment-wide association study (EWAS) using UK Biobank data identified several shared environmental factors across domains like health conditions, biological sample parameters, body index, and attendance availability. A shared etiology landscape for AD and AMD was then generated by integrating these novel findings with existing knowledge.
